Well, it looks like I may be doing a week long vacation in Cayo Coco! Sort of a last minute decision, just to laze around the beach and relax.

Having said that, what are my best options for buying boxes of cigars? Short of flying to Havana for a day, are there any reputable shops on the Island?

There is an LCDT at the Melia Cayo Guillermo resort where I have purchased boxes, this shop is a few mile up the road from the resorts at Coyo Coco. If he is still there, the managers name is Ramone,
I have been in a couple of the shops at other resorts in the area but cannot vouch for them, they were Caracol shops, not controlled by the LCDH.
IMO don't buy singles or boxes from anywhere except from the shop at the Melia.

How was the selection at the Melia? Lots of different vitolas?
How far is it from the Tryp? Walking distance I hope.

I went there in early September. Never again. The mosquitoes after sundown completely prevented any outdoor activity without repellent.

As for cigars. Tiendas Caracol used to deliver preordered cigars to a a couple of locations. No more. BTW Caracol run the LCDH brand of stores. So do Universelo.

The Tryp is not that close to the Melia, at least 8 miles, better take a cab or use a bicycle if you need the exercise
The selection varies. When I was there a couple of years ago they just got a shipment in to the store, must have been 500 boxes with a good selection
